T5 Exonuclease degrades DNA in the 5 to 3 direction. T5 Exonuclease is able to initiate nucleotide removal from the 5 termini or at gaps and nicks of linear or circular dsDNA. However, the enzyme does not degrade supercoiled dsDNA. T5 Exonuclease also has ssDNA endonuclease activity.

During tissue dissociation, parts of the cells are lysed resulting in a release of DNA. Monomolecular DNA may cause clumping of cells. Addition of DNase I to the dissociation buffer leads to a degradation of this extracellular DNA, therby avoiding the loss of cells from undesired clumping.

Vector Laboratories ImmPACT™ VECTOR Red Alkaline Phosphatase (AP) Substrate Kit 120 ml
ImmPACT™ Vector® Red AP Substrate Kit produces a bright red reaction product that is crisper and 2-4 times more sensitive than the original VECTOR Red Substrate. Sections stained with ImmPACT Vector Red substrate can be dehydrated, cleared, and permanently mounted. This chromogen can be used with other alkaline phosphatase or peroxidase substrates for multiple label applications. Because of its stability under ISH conditions, VECTOR Red can be used in IHC/ISH double labeling applications.
